AMIFLEX® is a nutritional supplement for maintaining healthy joints and to support flexible movement in humans. Amiflex® is also used to meet the increased need for nutrients of the musculoskeletal system at high load.
The building blocks for cartilage cells have a long way to go before they can be absorbed by the cells. Just below the inner coating (synovial membrane) of the joint capsules lies a loose connective tissue with many capillaries and lymph vessels. The nutrients from the capillaries are released into the synovial fluid in the synovial cavity. Regular joint movements mix the synovial fluid and the nutrients reach the cartilage surface. Diffusion takes place due to the concentration gradient and the nutrients eventually reach the cartilage cells.
Joint cartilage is mainly made up of three elements that offer flexibility, elasticity and firmness: proteoglycans to attract and retain water, collagen to fixate the proteoglycans and cartilage cells. The cartilage cells play an important role in maintaining the integrity of the tissue by means of the synthesis of collagen, proteoglycans and enzymes of the cartilage metabolism.
AMIFLEX® contains undenatured or native type II collagen in low doses, allowing to interact with the immune system. When cartilage is damaged, fragments (foreign to the body) are released into the synovial fluid. The immune system responds (immune response) with an inflammatory reaction in which antibodies against type II collagen are formed. Supplementation of undenatured type II collagen in low doses (milligrammes) desensitises the immune system so that further degeneration of the collagen fibres is prevented. When taken orally, specific parts (epitopes) of the intact helix structure are recognised by the immune system near the Peyer's patch in the last part of the small intestinal wall (ileum). After contact between undenatured type II collagen and the Peyer's patch, regulatory T-cells (Tregs) in the intestinal mucosa suppress the defence mechanism. The specifically formed regulatory T-cells migrate to the joint and reduce the inflammatory response. This working mechanism is called oral tolerance.
AMIFLEX® contains pomegranate extract that protects the cartilage cells. The skin of the pomegranate is rich in punicalagins. These water-soluble polyphenols with low molecular weight are powerful natural antioxidants that help combat oxidative stress. Pomegranate extract is valuable to maintain the integrity and function of the joints.
AMIFLEX® contains the free amino acids L-glycine, L-proline, S-adenosylmethionine, L-alanine and L-lysine for the synthesis of collagen. Collagen is a structural protein. It makes up a third of the mass of bone tissue and half of the dry weight of cartilage. Collagen ensures resilience.
AMIFLEX® contains glucosamine as a precursor of the glycosaminoglycans chondroitine sulfate and hyaluronic acid, an important component of the joint fluid and the cartilage matrix. Glycosaminoglycans (GAGs) are bound to a core protein to form proteoglycans. Proteoglycans bind water molecules, which results in strength and shock absorption. The viscosity of the joint fluid is primarily dependent on the concentration of hyaluronic acid. Glucosamine is a small molecule which can easily pass through the membranes of the capillaries to arrive in the joint fluid.
AMIFLEX® contains the cofactors vitamin C, vitamin B6 and manganese. Vitamin C contributes to the synthesis of collagen. Vitamin B6 contributes to the formation of interconnections and bridges (cross-links) between the collagen fibres. Manganese contributes to the polymerisation of glucosamine.
1 tablet AMIFLEX® contains the ingredients 1.60 mg undenatured or native type II collagen, 750 mg glucosamine sulfate (2KCL), 300 mg pomegranate extract (Punica granatum L.), 100 mg L-glycine, 100 mg L-proline, 50 mg S-adenosylmethionine, 35 mg L-alanine, 25 mg L-lysine, 90 mg vitamin C (L-ascorbic acid, 82% RDA*), 3 mg vitamin B6 (pyridoxine HCL, 100% RDA*), 1.50 mg manganese sulfate (15% RDA*), 100 mg lecithin (phosphatidylcholine, E322), 50 mg silicon dioxide (anti-caking agent, E551), 30 mg magnesium stearate (anti-caking agent, E572), 50 mg stearic acid (anti-caking agent), 40 mg hydroxypropyl cellulose (stabiliser, E463), 6 mg sodium carboxymethyl cellulose (stabiliser, E466), 335 mg microcrystalline cellulose (bulking agent, E460i), 40 mg hypromellose (gelling agent, E464) and 2 mg glycerin (emulsifier, E422).
